Summary of INN Addendum 22.554: New Stems and Amendments
This document, dated 12/10/2022, is an addendum to the INN Working Document WHO/EMP/RHT/TSN/2018.1, which outlines the selection of International Nonproprietary Names (INN) for pharmaceutical substances. It represents the cumulative additions of stems approved since the original publication in 2018.
New Stems Added
New stems categorize pharmaceutical substances by therapeutic area or mechanism:
- Examples: Lists include "-adant" for adenosine receptor antagonists (e.g., ciforadenant, etrumadenant), "-bep" for engineered proteins (e.g., ensovibep), "-bactam" for β-lactamase inhibitors (e.g., ledaborbactam, vaborbactam), and many others across categories like CFTR modulators ("-caftor"), calcium-sensing receptor agonists ("-calcet"), and monoclonal antibodies (e.g., "-mab" stems). Each stem entry includes associated INN-approved names with reference numbers.
Amendments to Existing Stems
Definitions were updated for certain stems to broaden their scope:
- Eridine stem expanded from "analgesics, pethidine derivatives" to include "µ-opioid receptor agonists."
- Kisant stem changed from "histamine H receptor antagonists" to "inverse agonists."
- Other updates include refinements to definitions for stems like ilide, imus, (o)nidine, netant, prim, and renone, aligning with specific therapeutic classifications.
Changes in Nomenclature for Monoclonal Antibodies
- The infix "-gromab" was modified for monoclonal antibodies targeting skeletal muscle growth factors.
- The "-ki-" infix was introduced for antibodies targeting interleukins or interleukin receptors.
Reference
A new monoclonal antibody naming scheme adopted at the 73rd INN Consultation is available at WHO website.
