Clinical Cancer Research: Trial Data Updates
Ziftomenib for Acute Myelogenous Leukemia (AML)
A phase Ib/II study (KOMET-001) evaluating ziftomenib monotherapy in combination with venetoclax showed promising clinical responses. In 112 patients, the overall response rate (ORR) was 35% (39/112) with a complete response/complete response with partial hematological recovery (CR/CRh) rate of 25% (28/112). Study findings demonstrated deep and durable responses in relapsed/refractory NPM1-mutant AML cases, with limited myelosuppression and a low rate of treatment discontinuation. Ziftomenib is well-tolerated with an overall 3% discontinuation rate.
Petosemtamab for Head and Neck Cancer
Updated data from a phase I/II trial (MCLA-158) combining petosemtamab and pembrolizumab in recurrent/metastatic head and neck squamous cell carcinoma (HNSCC) showed a high objective response rate (ORR) of 63% in 43 evaluable patients. The trial demonstrated a durable response with a median duration of response not reached at the time of analysis. The drug combination with immunotherapy is being investigated for its potential in improving outcomes in high-risk patient populations.
Bavencio for Bladder Cancer
Newtop-line results from the JAVELIN Bladder Medley phase II trial combining avelumab (Bavencio) with sacituzumab govitecan showed a significant improvement in progression-free survival (PFS) compared to avelumab monotherapy. The median PFS for the combination was 11.17 months versus 3.75 months. Further investigation and ongoing studies are expected to validate and expand these findings.
